How to use / operate the system

  • How long can you cell culture?

    It is depending on the imaging room condition but as culture reference, the primary cell can live more than 2-3 days or more. (refer following culture refence table)

  • My vessel is not listed on the Tokai Hit support vessel. May I use other vessel?

    We can offer customized holder depending on the vessel. Please contact Tokai Hit for its information.
    In the past, we had developed one holder to support 6pc of 35mm dish, multi-slide holder, etc.

  • Is it possible to change media during imaging with your chamber?

    Yes. We have add-on options called KS-Type 1 or KSX-Type 1. Please see add-on page for its information.

    We had made 18 access ports for tube access (2.0mm tube use) for media / drug delivery experiment to WSKM series in the past.

  • Is it possible to deliver the drug during imaging with your chamber?

    Yes. Please see add-on option KS-Type 2 or KSX-Type 2 for Drug Delivery.
    Without touching the chamber unit, the drug delivery possible.

  • How much is the water evaporation rate? When should be add/refill the water to the chamber?

    Depending on the room condition but approximately 0.5ml – 1.0ml water evaporate every hour.

    For long term cell culturing, we recommend to add pre-heated water to the chamber through water inlet by using syringe manually in the morning when you come to lab or evening before you leave lab.

  • Which controller is better STXG or STXF (G2F or F1)?

    We recommend to use STXG or G2F controller. The initial cost is more than STXF or F1 controller but running cost of gas is great.

    ① 100%CO2 gas is easier to cheaper and easier and the lead time is shorter.
    ② Gas consumption rate are different.
    Example: if you were using 47L gas cylinder and running the system for 24/7.
    With STXG or G2F controller, it will last more than 20 month. But same situation with STXF or F1 controller, it will only last 1 month.

  • During preliminary operation or imaging, there are condensation built on the Top Heater and/or vessel lids.

    Depending on the room condition or vessel designed, rarely you may experience condensation.

    Solution: require to adjust Setting Value of Heater(s).

    ① Use external sensor which is packed with our system (most of our system is equipped this sensor as standard) or if you do not have sensor prepare extern sensor with thermometer.

    ② Use scotch tape to secure the sensor end to the bottom center of the vessel (inner side) which you are having condensation issue.

    ③ Pour the water to the vessel (same amount that will be used during experiment).

    ④  Confirm all Heater Setting Value are set as Tokai Hit reference (default) value and leave the system for 40 minutes.

    ⑤ if you still find the condensation:
    a) Increase the Top Heater Setting Value by 1.0 degree C.
    b1) decrease the Setting Value of Bath Heater by 1.0 degree C.

    b2) If your system is not with Bath Heater, decrease the Setting Value of Stage Heater.

    ⑥ Leave 30 minutes and confirm the Thermometer indication. If the Thermometer temp. is higher/lower than your desired temp. then repeat the procedure ⑤.
    * if the temp. is lower than desired temp. instead of decreasing the Setting Value but increase it 1.0 degree C per time for both Heater.

    ⑦ After confirming the thermometer temp. is same as your desired temp. Record each heater Setting Value for future experiment.

  • How much is the gas pressure for the system?

    Please adjust the gas pressure 14.5psi / 0.1Mpa / 1.0kgf/cm2.

    If the gas pressure is less than above amount, the CO2 cannot supply to chamber unit properly. Please avoid to supply high gas pressure. In care of supplying the gas more than 17.5psi / 0.3Mpa / 3.0kgf/cm2 then it may damaged the integrated CO2 sensor.

  • How to clean the system?

    Do not use volatile material such as benzene or thinner for cleaning. Use of such materials will discolor and/or damage key device surface.

    Please clean the device with soft cloth using small amount of diluted neutral detergent. Do not use organic solvents.

In the case of system malfunction, damaged, etc.

  • The heater Process Value (PV) is not increasing to Setting Value (SV).

    The heater may have heater disconnection.
    Please contact Tokai Hit with your device serial number (it should be indicated on the connector of chamber unit heating cable).

    If the room temp. is too cold. The temperature is escaping to the stage. Please adjust your room temp. at 25 degree C.

  • Top Heater has glass breakage or crack.

    Contact Tokai Hit with serial number information.
    Depending on the model, you can purchase a spare Top Heater or if you could return the damaged Top Heater, we can offer repairing.

  • CO2 is unstable.

    Confirm the gas pressure is set as 14.5psi / 0.1Mpa / 1.0 kgf/cm2. In case the gas pressure is lower, it may not be able to supply proper CO2 concentration.

    If CO2 is still unstable with adjusted gas pressure as above. Please contact Tokai Hit with the system serial information. It should be indicated on the rear panel of controller.

About warranty

  • Is it fragile? What is the warranty?

    We do not hear the controller unit failure but since the chamber unit is applied “Glass Heater” to keep the clear vision during the imaging. This is the part you need to take extra care during the handling.

    Warranty is 12 month (18month from the shipping date) however glass breakage is not covered.

    The strengthen glass heater type ThermoPlate has 10 years glass breakage warranty.
